The present study was conducted to evaluate the effects of different doses of haloperidol (HP) on induction of oxidative stress in blood and liver cell degeneration in comparison with influences of HP pre-treatment on inflammatory process induced by intraperitoneal (IP) administration of lipopolysaccharide (LPS). One hundred twenty male albino Wistar rats were randomly divided into eight groups (15 in each), including: Control group, LPS group, three groups as HP administration in three divided doses (0.50, 1.00 and 2.00 mg kg-1), and three treatment groups that HP was administered in three doses (0.50, 1.00 and 2.00 mg kg-1) prior to LPS administration.Concentrations of malondialdehyde, activities of antioxidant enzymes including glutathione peroxidase, superoxide dismutase and also the levels of tumor necrosis factor-alpha and interleukin 1-beta were measured in blood and serum. In addition to liver histopathological changes evaluation, hepatic silent information regulator of transcription 1 (SIRT1) and phosphorylated-nuclear factor-κB (p-NF-κB) levels were quantitated. Our findings indicated that sole administration of HP (particularly higher doses) can induce oxidative stress in blood and cell degeneration in liver, while it can attenuate inflammatory process induced by LPS administration presumably via SIRT1 up-regulation and preventing the induction of p-NF-κB. The oxidative and degenerative effects of HP and its impact on inflammatory status were completely dose- dependent according to our results. The possible anti-inflammatory effects of HP may affect reparative mechanisms and hepatic cell degeneration. However, the influences of HP on immune system need further investigations and its higher doses should be administered cautiously especially in patients with immune system dysfunctions.

Gamma radiation are used in many medical and technical applications, however, it is one of the most dangerous kinds of radiation and can be harmful to the body. The present study was designed to clarify the protective effects of the selenium supplementation as selenium nanoparticle and selenite selenium in rat liver against Gamma irradiation with different intensities of 2.00 and 8.00 Gy. A total number of 45 healthy male Wistar rats were randomly divided into nine groups of five each. The radiation procedure was carried out in the Cobalt 60 equipment in Omid hospital, Urmia. The animals were simultaneously immobilized in a transparent acrylic plate and exposed to different intensities of 2.00 and 8.00 Gy radiations on day 7th and 14th of the experiment. After 72 hr after the last radiation, the animals were euthanized, and blood and liver tissue were collected. Histological analyses revealed the radiation-induced hepatic injury in rats, which included vacuolated cytoplasm, liver necrosis, fibrosis, and vascular lesions followed by a significant increase in alanine transaminase, alanine transaminase, alkaline phosphatase, and Gamma-glutamyl transferase. Selenium nanoparticles bear a more potent antioxidant effect in comparison with selenium selenite and can effectively protect the liver cell against Gamma radiation at a dose of 8.00 Gy.

Cyclophosphamide is a chemotherapy drug for the treatment of cancer. Chicken embryo amniotic fluid, vitamin C and coenzyme Q10 have anti-oxidant properties. Total of 70 adult female mice were selected and divided into seven groups. The first group that received 2 ml kg-1 of inactivated amniotic fluid subcutaneously. The second group treated with 75 mg kg-1of cyclophosphamide by intraperitoneal injection. Third to fifth groups received 1, 2, and 4 ml kg-1 of chicken embryo amniotic fluid, respectively. The sixth group received vitamin C at a dose of 0.2 mg g-1 of body weight by oral gavages. Seventh group received 10 mg kg-1 coenzyme Q10 intraperitoneally. All cyclophosphamide treated groups (3-7) received 75 mg kg-1 of cyclophosphamide intraperitoneal on day 22. The mice were euthanized on day 29 and ovarian tissue antioxidant enzymes including glutathione peroxidase, superoxide dismutase and catalase activities and malondialdehyde (MDA) were evaluated. Activities of above mentioned enzymes in treatment groups (3-7) was significantly higher than patient control group (2). The results also revealed that MDA levels were higher in the control group in comparison to other treatment groups. Therefore, it is concluded that the chick embryo amniotic fluid and coenzyme Q10 can compete with compounds like vitamin C in increasing the anti-oxidant level in ovarian tissue.

Chitosan bears numerous properties, such as biocompatibility, biodegradability and non-toxicity making it suitable for use in different biomedical fields. Zinc (Zn) is required for fibroblasts proliferation and collagen synthesis as essential elements of wound healing. Its nanoparticles are well known for their capability to enhance wound healing by cell adhesion and migration improvement through growth factors-mediated mechanisms. Poor blood supply and unique histological characteristics of tendon make its regeneration always slow. Also, adhesion formation between tendon and its surrounding tissues is another problem for neotendon to return to its normal structure and functional activities. In this study, a novel tubular scaffold of zinc oxide (ZnO) nanoparticles loaded chitosan has been fabricated for tendon repair. Experimental complete tenotomy of deep digital flexor tendon in a rabbit model was done and scaffolds were placed in the transected area after two ends suturing. After four and eight weeks, adhesion formation around the tendons and tissue reaction to the scaffolds were evaluated macroscopically. Inflammation, angiogenesis and collagen fibers arrangement were also analyzed in histopathological evaluations. After eight weeks, the scaffolds were absorbed completely, adhesions around the tendon were decreased and there was no sign of significant tissue reaction and/or infection in histopathological analyses. The reduced adhesion formation, improved gliding function and better histopathological characteristics suggest this scaffold application as a potential therapy in treatment of tendon acute injuries.

Blood selenium and trace minerals play an important role in animal’s health and production. The aims of this study were to determine selenium effect on blood glutathione peroxidase (GPX) activity, trace minerals and weight gain in lambs. Twelve female Makuei breed were studied for 63 days in groups of control, nanoselenium (NanoSe) and sodium selenite (NaSe). Mean concentrations of GPX, Cu and Fe in selenium supplemented groups were higher than in control group but the differences were not significant. Mean GPX and selenium was significant among the bleeding times, for Cu and Zn significant occasionally while not for weight gain. The percentages of weight gain in groups were 34.20, 38.90 and 36.30, respectively, which was not different. The individual comparison of parameters among groups showed differences for GPX, selenium and Cu. Correlations were observed between weight & Fe, weight & GPX, weight & selenium, Zn & Fe and GPX & selenium in NanoSe group. Weight gain showed negative correlations with Fe and positive correlation with GPX. In conclusion, selenium compounds increased GPX activity and selenium in which it was predominant in NanoSe than in NaSe group. Selenium compounds showed no effects on Cu, Zn and Fe but caused weight gain to increase. NanoSe revealed correlations between weight gain, GPX, Fe and selenium and was preferable to NaSe. Thus, the effect of NanoSe on reducing the oxidative stress and increased weight gain was acceptable and probably an option to NaSe administration in lambs.

In the present study, we investigated the effects of histidine and vitamin C (alone or in combination) treatments against isoproterenol (a β-adrenergic receptor agonist)-induced acute myocardial infarction in rats. We used propranolol (a β-adrenergic receptor blocker) to compare the results. Rats were given intraperitoneal injections of histidine (40 mg kg-1) and vitamin C (40 mg kg-1) alone and combined daily for 21 days. Propranolol (10 mg kg-1) was orally administered daily for 10 days (from day 11 to day 21). Myocardial infarction was induced by subcutaneous injections of 150 mg kg-1 of isoproterenol at an interval of 24 hr on days 20 and 21. Blood and tissue samples were taken for histopathological and biochemical evaluations following electrocardiography recording on day 21. Isoproterenol elevated ST segment, increased heart weight, heart rate, serum activities of aspartate transaminase, lactate dehydrogenase, creatine kinase-MB and heart tissue content of malondialdehyde, and decreased R wave amplitude and superoxide dismutase and catalase activities of heart tissue. Necrosis, edema and inflammatory cells infiltration were observed in myocardial tissue sections. Our results indicated that histidine and vitamin C alone, and especially in combination prevent isoproterenol-induced cardiotoxicity and have similar protective effects with propranolol. Cardioprotective effects of histidine and vitamin C may be associated with their ability to reduce free radical-induced toxic effects.

The present study investigated the effects of E. coli lipopolysaccharide (LPS) induced mastitis model in rat on the activity of antioxidant enzyme systems. To achieve this purpose, E. coli LPS were infused into the mammary gland of 12 rats 72 hr postpartum and compared with 12 rats in control group infused intramammary placebo sterile pyrogene – free, physiological saline. The antioxidant activities of the enzymes, superoxide dismutase, glutathione peroxidase, and catalase together with malondialdehyde (MDA) level were assayed in blood serum, milk and mammary tissue. Results obtained showed that, the antioxidant enzyme activities in milk, blood serum and mammary tissue were significantly decreased while the level of MDA, the indicator of lipid peroxidation were significantly increased following intramammary inoculation of LPS compared to the control animals. Histopathological examination also revealed the infiltration of inflammatory cells in mammary tissue and disruption of acinar structure and acinar lumina in mastitic rats. The results indicated that LPS-induced mastitis could alter antioxidant enzymes and increase lipid peroxidation.

Measurement of serum creatinine (Cr) and blood urea nitrogen (BUN) are used as indicators of glomerular filtration rate. The increased levels of these biomarkers are usually detectable at advanced stages of kidney complications. The aim of this study was to find the interrelationship of beta-2 microglobulin (β2M), BUN and Cr in streptozotocin (STZ)-induced diabetes mellitus in rabbits. Diabetes was induced by a single intraperitoneal (IP) injection of 65 mg kg-1 of STZ in rabbits. The levels of serum insulin, glucose and three above mentioned biomarkers were measured one day before (day -1) and on days 1-3 after injection of STZ and continued weekly to the end of the experiment (12 weeks). A statistically significant increase of serum β2M, BUN, Cr and glucose levels, and a significant decrease of insulin levels were observed in diabetic animals. However, β2M levels increased as early as one day after STZ injection compared to Cr and BUN that elevated at day two, suggesting a probable diagnostic advantage of β2M over currently used biomarkers in diabetic related kidney complications.

This prospective study aimed to compare the intraosseous (IO) and intravenous (IV) effects of propofol on selected blood parameters and physiological variables during general anesthesia in rabbits. Thirty New Zealand White rabbits were studied. Six rabbits received IV propofol (group 1) and another 6 rabbits, were injected propofol intraosseously (Group 2) for 30 minutes (experimental groups). Rabbits of the third and fourth groups received IV and IO normal saline at the same volume given to the experimental groups, respectively. In the fifth group IO cannulation was performed but neither propofol nor normal saline were administered. Blood profiles were assayed before induction and after recovery of anesthesia. Heart and respiratory rates, rectal temperature, saturation of peripheral oxygen and mean arterial blood pressure were recorded. Heart rate increased significantly 1 to 5 minutes after induction of anesthesia in experimental groups (P < 0.05). Although mean arterial blood pressure decreased significantly from baseline, values remained above 60 mm Hg (P < 0.05). Respiratory rate decreased significantly in experimental groups, but remained higher in group 2 (P < 0.05). The lymphocyte count decreased significantly in group 1 (P < 0.05). The concentration of alkaline phosphatase in all rabbits, aspartate aminotransferase and gamma-glutamyl transferase in the first group and gamma-glutamyl transferase in the third group increased significantly (P < 0.05). Total bilirubin decreased significantly in group 2 (P < 0.05). All measured values remained within normal limits. Based on the least significant physiological, hematological and biochemical effects, the IO injection of propofol appears to be safe and suitable method of anesthesia in rabbits with limited vascular access.

The material was incubated with sheep ruminal fluid and the experimental design was a completely randomized design. Cumulative gas production was recorded at 0, 2, 4, 6, 8, 12, 16, 24, 36, 48, 72 and 96 hours of incubation. Conventional in vitro OMD was determined after 48 hours incubation in acid and pepsin solutions. Samples for DaisyII OMD prepared according to the operating instructions supplied by ANKOM® Tech. Co., Fairport, USA. Compared to in vitro dry matter digestibility (IVDMD), DaisyII and gas production techniques overestimated (P < 0.05) OMD. The addition of EO and MON reduced (P < 0.05) the organic matter (OM), neutral detergent fiber (NDF), acid-detergent fiber (ADF) digestion, and gas production volume. The potential of gas production and rate of gas production for EO and MON were the lowest; however, these variables were higher for GAR supplemented groups. It was concluded that raw garlic could be of great interest for its usage as a modulator of ruminal fermentation.

Babesia ovis plays an important role in causing anemia and kidney dysfunction in affected animals. There are few extensive studies about hematological and biochemical findings of small ruminants' babesiosis caused by B. ovis. The aim of this study was to evaluate the effect of babesiosis on some hematological and biochemical parameters in infected small ruminants with B. ovis. A total of 280 sheep and 122 goats from 40 herds were randomly examined for the presence of B. ovis in blood samples. Of 402 samples, 67 animals (16.7%) were positive for B. ovis of which 52 (18.5%) were sheep and 15 (12.2%) goats, respectively. The infected animals were divided into four subgroups according to parasitemia rates (<1%, 1%, 2%, and 3%). As a control group, 67 uninfected animals were also selected from the same farms. With increase in parasitemia rates, hemoglobin concentration (Hb), packed cell volume (PCV), red blood cells (RBCs), mean corpuscular volume (MCV) and mean corpuscular hemoglobin concentration (MCHC) significantly decreased (P < 0.05), while, total leukocyte count, number of lymphocyte, monocyte, neutrophil and eosinophil showed a significant increase (P < 0.05). Infected animals presented a significant elevation (P < 0.05) of total proteins and significantly lower level (P < 0.05) of albumin compared to non-infected animals. Significant elevation (P < 0.05) of BUN, creatinine, cholestrol, triglyceride, HDL and LDL level were found with parasitemia progression.

Seventy six high and low producer cows were selected to determine the composition of the blood and milk parameters, and their interrelationships to determine the indices which could be useful to improve the milk yield. The highest mean blood concentrations were found in high producer cows. Mean values for blood urea nitrogen (BUN), serum protein (SPtn), creatinine, triglycerides (TGs), cholesterol, and beta-hydroxybutyric acid (BHB) were 25.10 mg dL-1, 10.15 g dL-1, 0.81, 62.30, 177.10 and 0.16 mmol L-1, and for macro-minerals including SCa, SMg, serum in-organic phosphorus (SIP), SNa and SK were 3.85, 2.66, 4.63, 108.00 and 4.34 mmol L-1, respectively. The highest concentrations for milk parameters, were observed in the high producers, and were significant only for MCa, MIP and MMg. Mean values for milk urea nitrogen (MUN), milk protein (MPtn) and lactose were 19.90 mg dL-1, 0.39 g dL-1, and 4.12% and for macro-minerals, 13.24, 3.88, 11.03, 73.30 and 16.90 mmol L-1, respectively. There were significant positive correlations between the blood and milk parameters except for creatinine/BHB, TGs/cholesterol and MNa/MK which were not significant. The correlations between the blood parameters were greater than in the milk parameters. Creatinine and SPtn, MUN and MPtn were the main parameters in that the relationships between MPtn with BUN, SPtn and creatinine were more noticeable than others. The regression analysis showed that BUN with the SIP and creatinine, MPtn with the BUN and creatinine and MUN with the SIP and SMg were the appropriate parameters in improvement studies related to the milk yield. In conclusion, BUN, SPtn, MUN and MPtn concentrations are the most effective indices for predicting the preferred milk yield.

In order to determine iron and lead content of seminal plasma in water buffalo and to study their associations with the semen characteristics, 54 semen samples were collected from 10 buffalo bulls. The semen characteristics were evaluated; its iron and lead content were estimated by atomic absorption spectrophotometer. The iron and lead content of the seminal plasma (Mean ± SEM) was recorded as 40.68 ± 0.75 mg L-1 and 0.026 ± 0.008 mg L -1, respectively. The mean iron value was highly associated with sperm progressive motility, gross motility and viability, negatively with lead content, and had a negative association with semen volume. The mean lead value was highly negatively associated with sperm progressive motility, gross motility, viability and positively associated with sperm abnormal morphology. For further clarification of these associations, the results were categorized in three groups of excellent (Ex, > 90 % motile, n = 33), good (Go, 80-89 % motile, n = 15) and moderate (Mo, < 79 % motile, n = 6) according to their percentage of sperm motility. The mean progressive motility in Ex, Go and Mo group was 92.24 ± 0.51 %, 81.66 ± 0.62 %, and 71.66 ± 1.05 % respectively. The mean iron and lead values and their associations with other parameters in these groups are discussed. The results show that seminal plasma iron content is associated with the motility and viability of the spermatozoa after ejaculation, but its lead content has an adverse effect on these parameters.

Rabbits are widely used as laboratory animals for experimental surgery. Anesthesia of rabbits may present complications unless the method is easy to apply and safe to use. In present study, effects of different dosages of vitamin C on thiopental sodium induced anesthesia in 25 male New Zealand white rabbits were studied. In the animals that had not received vitamin C treatment before thiopental sodium induced general anesthesia, return mean time of front limb pedal, corneal and ear pinch reflexes were 6.40 ± 1.67, 6.60 ± 2.96 and 8.00 ± 2.58 minutes, respectively. Pre-treatment of rabbits with 30 and 240 mg kg-1 (IV) of vitamin C followed by thiopental sodium 20 mg kg-1 (IV) resulted in significant (P < 0.05) increase in front limb pedal reflex return mean time to 13.00 ± 2.24 and ear pinch to 11.60 ± 4.16 minutes, respectively. There was also significant (P < 0.05) decrease in the heart rate following induction of anesthesia in the animals pre-treated with 30 and 90 mg kg-1 (IV) vitamin C and no change in the animals pre-treated with 240 mg kg-1 (IV) vitamin C. Serum analysis indicated a significant (P < 0.05) increase in blood glucose. These results suggest that premedication of rabbits with vitamin C despite potentiating of thiopental sodium anesthesia in rabbits is not dose dependent.

In order to determine calcium (Ca), magnesium (Mg) content and total antioxidant capacity (TAC) of seminal plasma in buffalo and to study their associations with the semen characteristics, 54 semen samples were collected from 10 buffalo bulls; semen quality was evaluated, seminal plasma was then harvested by centrifugation and its Ca and Mg content were estimated and its TAC determined. The Ca and Mg content of the seminal plasma (Mean ± SEM) were recorded as 22.36 ± 0.52 mg dl-1 and 11.94 ± 0.36 mg dl-1 respectively, while, its mean TAC value was 1.50 ± 0.02 mmol L-1. The mean Ca value was highly associated with sperm progressive motility, gross motility, viability (P = 0.000 for all), negatively with semen volume (P = 0.01), and with Mg and TAC values (P = 0.000 for both). The mean Mg values was highly associated with sperm progressive motility, gross motility and viability and seminal plasma Ca and TAC (P = 0.000 for all) and negatively associated with semen volume (P = 0.014).
