The present study aimed to investigate the protective effect of silymarin on maternal cadmium toxicity complications in the kidney of neonatal rats. Forty adults Wistar female rats were selected and placed with male rats for copulation. The pregnant animals were randomly divided into five groups (n = 8) including control, sham, silymarin, cadmium, and silymarin + cadmium. The animals received 400 mg L-1 cadmium and 100 mg kg-1 silymarin (sub-cutaneously, three days per week, three weeks). Two-day neonates were dissected and their right kidneys were fixed in 10.00% buffered formalin solution and processed by standard paraffin embedding. Tissue sections were stained by hematoxylin and eosin and analyzed histologically and stereologically. The data were statistically analyzed by SPSS using a one-way ANOVA test and Tukey's post-hoc. The results showed that silymarin significantly increased the neonatal rats' weight compared to the control group. Cadmium significantly decreased the weight of neonatal rats' kidneys. The results of histological studies indicated that cadmium caused subacute glomerulosclerosis, severe damage to urinary tubules such as tubular necrosis, and severe hyperemia in the medulla, but silymarin could preserve these complications. Stereological results revealed that cadmium decreased the total volume of kidney, medulla, and proximal and distal tubules and increased interstitial tissue and indicated the protective effects of silymarin on maternal cadmium toxicity complications in the kidney tissue of neonatal rats. It can be concluded that the administration of silymarin during pregnancy may be used as a useful and effective way of protecting the maternal cadmium toxicity complications in the kidney tissue of neonatal rats.

Osteoporosis is determined by decreased bone strength that increases the threat of fractures. The aim of this study was to evaluate the effects of pentoxifylline (PTX) and alendronate (ALN), on the stereological parameters, and gene expression in callus of fracture in an experimental rat model of ovariectomy-induced osteoporosis (OVX). The OVX was induced in 90 female rats. Fourteen weeks later, a complete fracture on the right femur was made. Rats were divided into five groups: 1) control: no treatment; 2) sham: received daily distilled water; 3) daily 3.00 mg kg-1 ALN subcutaneously (SC); 4) daily 200 mg kg-1 PTX (SC) and 5) daily PTX (SC) + ALN (same doses). The osteoclast count was significantly lower in all treatment groups, at 21 and 56 days post-surgery, compared to the control and sham groups. The PTX significantly increased total callus volume at 21 and 56 days post-surgery, compared to the other groups. The PTX+ALN treatment significantly increased both cortical bone volume on day 21, and osteocyte and osteoblast numbers on day 56, compared to the control and sham groups. It can be concluded that PTX and ALN have antiresorptive effects, in OVX rats. Also, PTX has increased the extracellular matrix on both 21 and 56 days after surgery, compared to the other groups. PTX+ALN elevated cortical bone volume on day 21, and osteocyte and osteoblast numbers compared to the control and sham groups on day 56.

Pomegranate (Punica granatum L.) has been used widely in the traditional medicine of various civilizations for more than 5000 years. The pomegranate tree has several parts; each part has useful medicinal effects. Previous studies have demonstrated the antibacterial, antioxidant, and anti-inflammatory properties of pomegranate. The aim of the present study was to determine whether administration of pomegranate extract could result in morphometric changes in the kidneys of rats. Eighteen male rats (180-200 g) were divided into three groups that received either: G1, distilled water; G2, 250 mg kg-1 pomegranate extract; and G3, 500 mg kg-1 pomegranate extract via oral gavages daily for eight weeks. At the end of eight weeks, the rats were euthanized and their kidneys were removed and processed for morphometric analyses. In rats received pomegranate extract, the kidney weight, kidney weight/body weight ratio, cortex volume and glomerular volume were increased (p < 0.05), while, medulla volume and the number of glomeruli per kidney did not change. No pathological lesions were observed in the kidney. Therefore, pomegranate hydro-alcoholic extract at doses of 250 and 500 (mg kg-1) increased the volume of some parts of the kidney; however, it did not cause any pathological changes in the kidney.

The aim of the present study was stereological evaluation of testes of azoospermic animal model using busulfan in rat. Three groups of male adult rats were used in this study. The first group was injected by single dose of busulfan (10 mg kg-1) and their testes were removed on day 35 post injection. The second group received double doses of busulfan with 21 days interval and their testes were removed on day 35 after the second injection. The testes of the third group were removed without busulfan therapy. In 10 circular transverse sections of tubules stained with hematoxylin-eosin, stereological parameters were measured. The testes were rated for its spermatogenic potential on a modified spermatogenic scale of 0 to 6. Cellular (germinal epithelium) diameter and area of the seminiferous tubules, total diameter and cross sectional area of the tubules of the seminiferous tubules in rats that received double doses of busulfan were less than the rats in single dose of busulfan and control groups (p < 0.05). Spermatogenesis index of seminiferous tubules in rats receiving two doses of busulfan was less than the rats received one dose of busulfan (p < 0.001) and the index of both treatment groups were less than the control group (p < 0.001). In conclusion, two doses of busulfan injection with 21 days interval produced an appropriate experimental model of induced azoospermia with comparable stereological indices of seminiferous tubules in rat.

In pancreatic survey, quantitative and morphological characteristics could be fundamental variables for the evaluation of some structures. The aim of this study was the application of stereological methods for estimation of quantitative parameters and morphological evaluation of the pancreas in chickens. Ten adult male and ten adult female native chickens were used in this study. Routine tissue processing was carried out for all samples. The sections were stained with hematoxylin and eosin and Gomori's aldehyde-fuchsin. Stereological examinations were performed according to systemic uniform random sampling as well as Cavalieri and point counting method. The morphological results showed that in both sexes, pancreas of chickens was composed of four main pancreatic lobes and contained two distinct islet types. Alpha islets consisted of alpha and a few delta cells and beta islets contained beta and seldom delta cells. Stereological results showed that in both sexes, there were significantly distinctive regional differences in the volume of islet and the numerical density of cells among some lobes with highest values in third and splenic lobes, respectively (p < 0.05). The nuclear volume was lowest in ventral lobe either males (80.60 ± 33.50) or females (84.20 ± 44.10). Beside, in the islets diameter of splenic lobe, there were significant differences in males as compared with females (p < 0.05). In splenic lobe, a significant difference was observed in the nuclear diameter in males than to females (p < 0.05). The results indicated that although there were not morphological changes between sexes in pancreatic lobes, however, some stereological parameters could be affected by sex.
