The objective of the present study was to evaluate the efficacy of a porcine circovirus type 2 (PCV2) double vaccination in weaning piglets reared for gilts under field conditions. The study was conducted at a Greek farrow-to-finish conventional pig farm with a previous history of PCV2 infections. The trial included 96 female piglets at 21 days of age, which were equally allocated to two different study groups. Piglets of the group-1 received a single PCV2 vaccination at 21 days of age, while piglets of the group-2 were double vaccinated against PCV2, at 21 and 42 days of age. The results indicated that the piglets of group-2 had better growth performance, as they showed higher body weight (BW) and average daily weight gain (ADWG). In addition, ELISA tests showed that the double- vaccinated piglets presented a better humoral response against PCV2, as higher levels of IgG antibodies were detected in them than the piglets of the group-1. In conclusion, the current results suggested that a double PCV2 vaccination of piglets, reared for gilts, on a PCV2-affected farm could lead to higher protection against the virus.

Antibodies are essential tools of biomedical and biochemical researches. Polyclonal antibodies are produced against different epitopes of antigens. Purified F(ab')2 can be used for animal’s immunization to produce polyclonal antibodies. Human immunoglobulin G (IgG) was purified by ion exchange chromatography method. In all stages verification method of the purified antibodies was sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Purified IgG was digested by pepsin enzyme and F(ab')2 fragment was purified by gel filtration separation method. For production of polyclonal antibody, rabbit was immunized by purified F(ab')2 and antibody production was investigated by enzyme-linked immunosorbent assay. Purified anti-IgG F(ab')2 was conjugated with fluorescein isothiocyanate. Ion exchange chromatography purification yielded 38 mg of human IgG antibody. The results of SDS-PAGE in reduced and non-reduced conditions showed bands with 25-30 kDa molecular weight (MW) and 50-kDa respectively and a distinct band with 150 kDa MW. The results of non-reduced SDS-PAGE for determining the purity of F(ab')2 fragment showed one band in 90 kDa and a band in 150 kDa MW position. Purification by Ion exchange chromatography method resulted about 12 mg rabbit polyclonal antibody. Flow cytometry showed generated polyclonal antibody had an acceptable activity compared to commercial antibody. Taking together, purified IgG F(ab')2 and polyclonal anti-IgG F(ab')2 are useful tools in biomedical and biochemical researches and diagnostic kits.

To investigate the IgG content and its variations in uterine fluid (UF) during the estrous cycle of the cow and to compare them with those of the blood serum (S), six pairs of serum and UF samples for each phase of the cycle selected out of 240 bovine genital tracts and blood samples were collected from Urmia abattoir. The UF samples were collected by gentle scraping of the endometrium using a curette after uterine incision and their IgG content and those of the serum were measured by single radial immuno-diffusion (SRID) assay. Serum IgG values (Mean ± SEM) were generally higher than the UF values throughout the cycle except for di-estrus (S: 38.50 ± 0.90, UF: 51.60 ± 2.10 mg mL-1), in which the highest values were observed in UF samples. In met-estrus the difference was not significant (S: 34.80 ± 1.80mg mL-1, UF: 30.80 ± 5.20 mg mL-1), however, in estrus the mean UF IgG value (12.50 ± 1.10 mg mL-1) was lower than that of the serum (31.30 ± 1.20 mg mL-1). In pro-estrus, the lowest values (S: 27.80 ± 1.30 mg mL-1, UF: 9.10 ± 1.50 mg mL-1) were obtained. The results showed a lower IgG values in the bovine UF than those of the serum in the follicular phase of the cycle, while in di-estrus the UF IgG content was the highest, suggesting some IgG production in the uterus at this phase.
