Limited information exists about the relationship of adipose tissue with inflammation, oxidative stress, and energy metabolism during the transition period in dairy cows. The objective of this study was to assess the changes and relation of some adipokines, cytokines, oxidative biomarkers, and serum biochemical parameters related to energy balance (EB) in cows during the transition period. Thirty multiparous Holstein cows were selected based on estimated parturition date, and blood samples were collected from jugular vein on one-week prepartum and one and three weeks postpartum and used to measure the parameters. The serum levels of beta-hydroxybutyric acid (BHB), non-esterified fatty acid, cholesterol, high-density lipoprotein (HDL), aspartate aminotransferase, and total antioxidant capacity increased significantly, and glucose, urea, triglyceride (TG), and low-density lipoprotein (LDL) decreased significantly after parturition. The serum values of adiponectin, resistin, leptin, and cytokines including interleukin 6 (IL-6) and tumor necrosis factor (TNF)-α were not changed significantly during the experiment. The results of the Pearson correlation revealed a significant negative correlation between BHB with glucose, albumin, cholesterol, HDL, LDL, and a positive correlation with TG and malondialdehyde. Also, there was a significant direct correlation between insulin and leptin, adiponectin, resistin, IL-6 and TNF-α in the whole experiment period. These emphasize the difficulty of dairy cows to manage the energy requirements during the transition period. It can be stated that adipokines and cytokines may have an essential role in the metabolic status in this period, and control of their production and, or secretion could be helpful in EB during the transition period.

In this study, it was aimed to investigate the association between inflammatory reaction of tumoral microenvironments with interleukin responses in ovine pulmonary adenocarcinomas (OPAs). Material of the study consisted of 26 sheep lung tissue samples being brought to the Pathology Department for routine diagnosis. Cases were collected between years 2009 - 2021; pre-diagnosis was based on clinical symptoms, anamnesis and gross lesion of the lungs. These tissues were designated in two groups as control (n = 6) and OPA (n = 20) groups. Choice of immunohistochemical staining was avidin-biotin peroxidase method. Reverse transcription polymerase chain reaction (RT-PCR) was used to confirm Jaagsiekte sheep retrovirus from paraffin-embedded tissues. On gross examination of OPAs, lesions seen were mostly in the caudal lobes of the lung, 1.00 – 2.00 cm in diameter as gray-white consolidated foci and in microscopic observation, tumor cells showed acinar, papillary or mixed growths. No expressions of interleukin (2 and 8) were observed in the control group. All OPAs cases were positive for interleukins (2 and 8) expressions. A total of eight tissue samples were detected as positives through RT-PCR. In conclusion, in this study, it was determined that interleukin-2 and interleukin-8 were produced from tumor microenvironment elements, especially tumor-associated macrophages, and these interleukins showed pro-inflammatory effects. Interleukins and the inflammatory reaction may promote the development of OPA.

The study was aimed to evaluate the effects of hydro-ethanol extract Zataria multiflora on the brain tissue oxidative damage, and hippocampal interleukin-6 (IL-6) as well as learning and memory capacity in lipopolysaccharide (LPS) - challenged rats. The rats were randomized into five groups as follow: Control group: Rats were treated with saline, LPS group: Rats were treated with LPS 1.00 mg kg-1, ZM50, ZM100 and ZM200 groups in which the rats were treated with Z. multiflora extract (50.00, 100 or 200 mg kg-1 per day, respectively). The treatments including extract or vehicle were administered intraperitoneally and given three days before the behavioral tests and were continued within a6-day behavioral experiment. Injection of LPS was daily done before the behavioral tests. Finally, the brains were collected for biochemical evaluations. Although LPS administration prolonged the latency in Morris water maze and shortened the latency to enter the dark chamber in passive avoidance test, ZM extract restored these changes to approach control group values. Also, LPS increased IL-6, malondialdehyde (MDA) and nitric oxide (NO) metabolites levels and lowered thiol, superoxide dismutase (SOD) and catalase (CAT) levels in the brain, however, Z. multiflora extract reduced IL-6, MDA and NO metabolites concentrations, but increased thiol content, SOD, and CAT levels. The results of this study showed that Z. multiflora ameliorated learning and memory dysfunction in LPS - challenged rats by alleviating of inflammatory responses and brain tissue oxidative damage.

Uveitis is a major cause of vision loss. Methotrexate (MTX) has been widely used in uveitis due to its relatively safe profile. The purpose of this study was to evaluate the effects of two different dosages of MTX via intra-vitreal administration for treatment of endotoxin induced uveitis (EIU) in an experimental model. Thirty-five healthy rabbits were randomly divided into four groups and all animals were tolerated intra-vitreal injections. The first group received normal saline (NS), the second group received normal saline plus Salmonella typhimurium lipopolysaccharide endotoxin (LPS), (NS+LPS), the third group received 400 μg MTX plus LPS (LPS+MTX 400) and the fourth group received 800 μg MTX plus LPS (LPS+MTX 800). Intra-ocular inflammation was evaluated by clinical examination scoring during 7 post-injection days and histopathological examination at the end of study. Kruskal-Wallis and Mann-Whitney U tests were used to compare the histopathological and clinical scores. According to the clinical examinations, all groups demonstrated higher uveitis score than group 1 on first post-injection day. Also, groups 2 and 3 showed greater uveitis score than group 4. On the third, fifth and seventh post-injection days, clinical uveitis score in groups 2, 3 and 4 was significantly higher than group 1. The mean histopathological inflammation intensity scores in groups 2, 3 and 4 were significantly higher than group 1. Single intra-vitreal injection of 400 μg and 800 μg of MTX did not show significant anti-inflammatory effects on EIU in rabbits.

Tendons are frequently targets of injury in sports and work. Whether nonsteroidal anti-inflammatory drugs (NSAIDs) have beneficial effects on tendon healing is still a matter of debate. This study was conducted to evaluate effects of flunixin meglumine (FM) on tendon healing after experimentally induced acute trauma. Twenty eight adult male New Zealand White rabbits were subjected to complete transection of deep digital flexor tendons followed by suture placement. Treatment group received intramuscular injection of FM for three days, and controls received placebo. Subsequently, cast immobilization was continued for two weeks. Animals were sacrificed four weeks after surgery and tissue samples were taken. The histological evaluations revealed improved structural characteristics of neotendon formation including fibrillar linearity, fibrillar continuity and neovascularization in treatment group compared to those of controls (p < 0.05). However, no significant differences were found between two groups in terms of epitenon thickness (p > 0.05). Mechanical evaluation revealed significant increase in load-related material properties including ultimate load, yield load, energy absorption and ultimate stress in treatment group compared to those of control group (p < 0.05). However, no statistically significant differences in terms of stiffness and ultimate strain were found (p > 0.05). The present study showed that intramuscular injection of FM resulted in improved structural and mechanical properties of tendon repairs and it could be an effective treatment for acute tendon injuries like severance and laceration.

In this study, the effects of separate and combined intraperitoneal (IP) injections of histidine and dexamethasone were investigated on local inflammation in rats. Local inflammation was induced by subcutaneous (SC) injection of histamine (100 μl, 0.1%) in ventral surface of right hind paw. The thickness of paw was measured at 30 min before and 30, 60, 90, 120, 150 and 180 min after injection of histamine, using a fine caliper. The number of neutrophils in paw tissue sections was counted 3 h after intraplantar (IPL) injection of histamine. The IPL injected histamine elicited an inflammatory response that was characterized by increase of paw thickness and by infiltration of neutrophils in paw tissues. IP injections of histidine at doses of 200 and 400 mg kg-1 and dexamethasone at a dose of 1 mg kg-1 significantly (P < 0.05) decreased both paw thickness and infiltration of neutrophils in paw tissues. In combined treatment, IP injection of histidine (200 mg kg-1) with dexamethasone (1 mg kg-1) produced a more documented response in comparison with histidine and dexamethasone used alone. The results suggested that histidine and dexamethasone have anti-inflammatory activities. Histidine potentiated the anti-inflammatory effect of dexamethasone in histamine-induced local inflammation.
