Protective effect of the bee bread on cadmium-induced testicular toxicity in rats

Abstract Cadmium (Cd) is a highly toxic environmental pollutant known to cause severe damage to the male reproductive system. This study aimed to investigate the protective effects of bee bread (BB), a natural product with anti-oxidant, anti-apoptotic, and anti-inflammatory properties, against Cd-induced testicular toxicity in male Wistar rats. A total number of 32 rats were divided into four groups, including control, BB (0.50 g kg-1), Cd (5.00 mg kg-1), and Cd + BB (5.00 mg kg-1 and 0.50 g kg-1, respectively) groups. Administrations via oral gavage were performed for 4 weeks. Semen analysis revealed significant reductions in sperm motility and density along with increases in abnormal and dead sperm ratios in the Cd and Cd + BB groups compared to controls. Histopathological examination showed severe degeneration and desquamation of germ cells, tubular atrophy, and a decrease in spermatozoa in the Cd-treated groups. Polymerase chain reaction analysis indicated up-regulation of apoptotic markers (caspase-3, -8, and -9) and oxidative stress enzymes (catalase and superoxide dismutase) in the Cd group, signifying disrupted testicular function. The BB administration partially mitigated Cd-induced damage as evidenced by less severe histopathological changes and moderated gene expression alterations. However, the protective effects of BB were not sufficient to completely counteract the toxic impact of Cd. The present study concluded that while BB had potential in reducing Cd-induced testicular toxicity, its protective efficacy was limited, warranting further research to explore its therapeutic potential in combination with other protective agents.
Determination of the effect of Tarantula cubensis alcoholic extract on cadmium embryotoxicity

Abstract Tarantula cubensis alcoholic extract (TCAE) is a homeopathic product used in the veterinary field. This study aimed to determine the effects of TCAE on cadmium (Cd) toxicity in the embryo. The study used 220 fertile, incubated chicken eggs divided into 11 equal groups on the 7th day of incubation. The groups comprised untreated and physiological saline control groups, a group with TCAE alone, four groups with varying doses of Cd alone and four groups with the same doses of Cd plus TCAE. At the end of the incubation period, the eggs were opened, kidney and liver tissue samples were taken for histopathology and the number of dead and living embryos were recorded. In the prsent study, the median lethal dose of Cd was determined to be 0.029 mg per egg and the median lethal dose of Cd plus TCAE was determined to be 0.020 mg per egg. The histopathological examinations determined that kidney and liver damage were increased when TCAE and Cd were administered together, that was higher than when Cd was given alone. Thus, TCAE, which had no toxic effect on the embryo when used alone, might increase the embryotoxic activity of Cd. However, more detailed studies are needed.
Silymarin protects the structure of kidney in the neonatal rats exposed to maternal cadmium toxicity: A stereological study

Abstract The present study aimed to investigate the protective effect of silymarin on maternal cadmium toxicity complications in the kidney of neonatal rats. Forty adults Wistar female rats were selected and placed with male rats for copulation. The pregnant animals were randomly divided into five groups (n = 8) including control, sham, silymarin, cadmium, and silymarin + cadmium. The animals received 400 mg L-1 cadmium and 100 mg kg-1 silymarin (sub-cutaneously, three days per week, three weeks). Two-day neonates were dissected and their right kidneys were fixed in 10.00% buffered formalin solution and processed by standard paraffin embedding. Tissue sections were stained by hematoxylin and eosin and analyzed histologically and stereologically. The data were statistically analyzed by SPSS using a one-way ANOVA test and Tukey's post-hoc. The results showed that silymarin significantly increased the neonatal rats' weight compared to the control group. Cadmium significantly decreased the weight of neonatal rats' kidneys. The results of histological studies indicated that cadmium caused subacute glomerulosclerosis, severe damage to urinary tubules such as tubular necrosis, and severe hyperemia in the medulla, but silymarin could preserve these complications. Stereological results revealed that cadmium decreased the total volume of kidney, medulla, and proximal and distal tubules and increased interstitial tissue and indicated the protective effects of silymarin on maternal cadmium toxicity complications in the kidney tissue of neonatal rats. It can be concluded that the administration of silymarin during pregnancy may be used as a useful and effective way of protecting the maternal cadmium toxicity complications in the kidney tissue of neonatal rats.
